Rice cold tolerance at the germination stage is important in Rio Grande do Sul (RS) where temperatures below 15°C prevent or reduce germination and plant establishment in early sowings. The present study aimed at identifying an adequate method for cold tolerance evaluation of the rice germination stage and at verifying the variability among 24 rice genotypes of different origins. Cold tolerance was evaluated in experiment I, germination under two conditions: 13°C for 28 days and 28°C for seven days, and in experiment II, germination under 28°C for 72 hours, 13°C for 96 hours and again 28°C for 72 hours. ABSTRACT Functional genomic analyses require intact RNA; however, Passiflora edulis leaves are rich in secondary metabolites that interfere with RNA extraction primarily by promoting oxidative processes and by precipitating with nucleic acids. This study aimed to analyse three RNA extraction methods, Concert™ Plant RNA Reagent (Invitrogen, Carlsbad, CA, USA), TRIzol® Reagent (Invitrogen) and TRIzol® Reagent (Invitrogen)/ice -commercial products specifically designed to extract RNA, and to determine which method is the most effective for extracting RNA from the leaves of passion fruit plants. Cotton zoning based on sowing periods of lower risk in Parana state, Brazil BABT

Cotton is cultivated in the North and West of Parana State, southern Brazil, under conditions of climatic risk variable in space and time. Risks of temperature below 15ºC at the establishment period, daily average temperature below 20ºC at the stage of cotton boll opening, and soil water deficit for both plant establishment and flowering periods, were estimated to identify homogeneous zones with sowing periods of lower climatic risk. The time interval with adequate temperature, associated with minimum risks of the other factors and yield data from field experiments allowed the identification of seven distinct zones, with best sowing periods ranging from September 20 to November 20. Development of eggs and the planktonic stages of salmon lice (Lepeophtheirus salmonis) at low temperatures Naturalis

To verify if and to what extent egg and nauplii development of the salmon lice take place during winter, the development from egg to the copepodid stage at 2,3,4,5 and 10°C was examined. Newly extruded egg strings from a winter population of salmon lice were individually placed in 6 ml stagnant hatching systems. Initially, no significant differences in egg development time were found between these and larger aerated systems, though a tendency for less synchronised hatching of the total egg string was detected in the stagnant systems (difference< 12 h). In light versus dark conditions the time to hatching was significantly prolonged by darkness (10-15%). The electrical conductivity test has been used to evaluate seed vigour in many species, especially legumes, such as peas, beans and soybeans. The objective of this research was to evaluate the effect of temperature and storage period on the results of the electrical conductivity test and the mineral composition of the imbibition solution of bean seeds (Phaseolus vulgaris L.). Simulating the vernalization response of the "Snow Queen" lily (Lilium longiflorum Thunb.) Scientia Agricola

Vernalization is a process required by certain plant species, including lilies (Lilium spp.), to enter the reproductive phase, through an exposure to low, non-freezing temperatures. The objective of this study was to evaluate a nonlinear vernalization response function for the "Snow Queen" lily. An experiment was carried out in Santa Maria, RS, Brazil, to provide an independent data set to evaluate the performance of the model. Lily bulbs were vernalized at -0.5, 4.0, and 10ºC during two, four, six, and eight weeks. Simultaneous environmental manipulations in semi-perfusion cultures of CHO cells producing rh-tPA Electron. J. Biotechnol.

We evaluated the combined effect of decreasing the temperature to a mild hypothermia range (34 and 31ºC) and switching to a slowly metabolizable carbon source (glucose substituted by galactose) on the growth and production of a recombinant human tissue plasminogen activator (rh-tPA) by Chinese hamster ovary cells in batch and semi-perfusion cultures. In batch cultures using glucose as a carbon source, decreasing the temperature caused a reduction in cell growth and an increase in specific productivity of rh-tPA of 32% at 34ºC and 55% at 31ºC, compared to cultures at 37ºC. Different egg conservation techniques were assessed under low temperature conditions to multiply the parasitoid of stink bug eggs Trissolcus basalis (Woll.) (Hymenoptera:Scelionidae). Masses of fresh Nezara viridula (L.) eggs were stored for 12 months in liquid nitrogen (-196ºC), wrapped in aluminum foil, in microcentrifuge tubes ("eppendorf") or in vacuum sealed double plastic bags (500 eggs/technique/month).
